Kisspeptin-10 is a ten-amino-acid fragment of the protein kisspeptin, also known as metastin. The protein is encoded by the KISS1 gene; the name metastin comes from the original research into that gene. Kisspeptin-10 is the shortest piece that still fits the KISS1 receptor (GPR54), a binding site on the cell membrane found among other places on the GnRH neurons in the hypothalamus. The end of the chain is amidated, which is characteristic of this family.
The switch here sits high up in the chain. Kisspeptin-10 is not a sex hormone itself and does not act on the gonads. It fits the KISS1 receptor of the GnRH neurons, and those sit one step before GnRH, the signal with which the pituitary releases LH and FSH. In the literature kisspeptin is therefore described as the top link of the hypothalamic-pituitary-gonadal axis. Other substances in this category act lower in that chain or via an entirely different route, such as PT-141 via the melanocortin receptors.
